Ramadan, Epolia, Zhanyan Fu, Gabriele Losi, Gregg E. Homanics, Joseph H. Neale, and Stefano Vicini. GABAA receptor 3 subunit deletion decreases 2/3 subunits and IPSC duration. J Neurophysiol 89: 128–134, 2003. 10.1152/jn.00700.2002. Deletion of the 3 subunit of the GABAA receptor produces severe behavioral deficits and epilepsy. GABAA receptor-mediated miniature inhibitory postsynaptic currents (mIPSCs) in cortical neurons in cultures from 3 / mice were significantly faster than those in 3 / mice and were more prolonged by zolpidem. Surface staining revealed that the number of 2/3, 2, and 3 (but not of 1) subunit-expressing neurons and the intensity of subunit clusters were significantly reduced in 3 / mice. Transfection of 3 / neurons with 3 cDNA restored 2/3, 2, and 3 subunits immunostaining and slowed mIPSCs decay. We show that the deletion of the 3 subunit causes the loss of a subset of GABAA receptors with 2 and 3 subunits while leaving a receptor population containing predominantly 1 subunit with fast spontaneous IPSC decay and increased zolpidem sensitivity.

Quantitative Analysis of GABAA Gamma Receptor Subunits in the Developing Embryonic Chick Forebrain
Objective(s) In this study we investigated the expression of GABAA receptor subunits during brain development. These receptors may change in the embryonic chick forebrain. Materials and Methodes The expression levels of four types of GABAA receptor gamma subunits (γ1, γ2, γ3 and γ4) were quantified in the embryonic chick forebrain at 32 hr, 3, 7, 14, and 20 days of incubation and day one aft...

First published January 16, 2004; 10.1152/japplphysiol.01264.2003.—Previously, we reported that the pre-Bötzinger complex (PBC) exhibited a dramatic reduction in cytochrome oxidase activity at postnatal da...
متن کاملDeletion of the 1 or 2 Subunit of GABAA Receptors Reduces Actions of Alcohol and Other Drugs
Enhancement of the activation of GABAA receptors is a common feature of many sedative and hypnotic drugs, and it is probable that the GABAA receptor complex is a molecular target for these drugs in the mammalian central nervous system. We set out to elucidate the role of the two predominant ( 1 and 2) subunits of GABAA receptor in sedative drug action by studying mice lacking these two subunits...
متن کاملGABAA Receptor Subunits in Rat Testis and Sperm
Background γ-Aminobutyric acid (GABA) is considered to be the predominant inhibitory neurotransmitter in mammalian central nervous systems (CNS). There are two major classes of GABA receptors: GABAARs and GABABRs. The GABAA receptor is derived from various subunits such as alpha1-alpha 6, beta1-beta 3, gamma1-gamma 4, delta, epsilon, pi, and rho1-3. Intensive research has been performed to und...
